1221 Tumor-immune cell interaction in melanoma progression arising from congenital melanocytic nevus analyzed by spatial transcriptomics

Tumor infiltrating immune cells (TIICs) interact with tumor to influence development and progression. However, how the interaction between melanoma TIICs affects invasion metastasis of in congenital melanocytic nevus (CMN) is largely unknown. We analyzed multiple samples including various regions different depths metastatic tissues from two patients identified differential gene expression profiles for using spatial transcriptomics. As depth increased, genes related cell exhaustion M2 macrophage polarization, such as UBE2T MDK, were increased cells. In addition, AXL, which promotes evasion, EPHA2, inhibits infiltration, lesions. macrophage, PRMT1, SERP1, RPS3, DAPK3,increased, while T cells, inhibitory signaling, LRMT1 PSTPIP1, according depth. this study, we heterogeneous composition phenotype CMN progressed metastasized.
